Police have confirmed a second man has died almost a week after a shooting outside Melbourne's Love Machine nightclub.

Richard Arow, 28, of Maidstone, was remembered on social media for his "everlasting smile", as police continued to investigate the people and motives behind the attack.

Police said he died last night in hospital.

Police believe at least five people were hit by shots fired from a car into the crowd outside Love Machine, at Prahran in Melbourne's inner south, about 3:10am on Sunday.

Aaron Khalid Osmani, 37, worked as a security guard at the venue and died from his injuries on Sunday.

Mr Arow was taken to hospital after being shot as he lined up to enter the venue.

Tributes to the Maidstone man posted on social media earlier this week included a message from the Westside Strikers Football Club, which remembered him as a premiership-winning teammate to many.

"Richard was held in high regard, his personality and everlasting smile is something we will always remember," it said.

Players planned to observe a minute's silence and wear black armbands at their next game, the club said.

"You were not only a leader on the park who gave 110 per cent, but a true gentleman off it. You will be sadly missed," West Strikers president Sam Raniolo commented.

Three other men were treated in hospital for non-life-threatening injuries following the incident and a young woman was taken to hospital after falling and hitting her head at the scene.

The anti-bikie Echo Taskforce and homicide squad are still working to understand what motivated the shooting, but believe it was targeted.

The venue's operators were cooperating with investigators, who were looking at a range of "different criminal entities", Victoria Police's Assistant Commissioner Tess Walsh said after the shooting.
