The Ringwood Bypass was closed during the afternoon peak between EastLink and Warrandyte Road.
The ramps from EastLink to the bypass were also closed, and Ringwood Street was shut between Loughnan Road and Eastland shopping centre.
All streets re-opened about 6.45pm on Monday.
VicRoads earlier said there was "significant congestion" on EastLink, the Maroondah Highway, Canterbury Road and Wantirna Road.
There have been 171 crash deaths on Victorian roads so far this year, up from 111 at the same time last year.
Police are urging anyone who witnessed the collision to contact Crime Stoppers on 1800 333 000.
